This census was taken about 40 years after the first census in Numbers 1, as the Israelites prepared to enter the Promised Land. Reuben was Jacob's firstborn son, so his descendants formed one of the twelve tribes of Israel. Interestingly, this tribe had actually decreased in number from the first census, where they numbered 46,500.
